    首先用碘酊染色顯微鏡體細(xì)胞計數(shù)法確定了100頭不同患病程度的奶牛奶樣的體細(xì)胞數(shù),然后采用叉指電極在0.01~100 kHz頻率范圍對奶樣進(jìn)行了交流阻抗測試。在提出該測試系統(tǒng)等效電路的基礎(chǔ)上用Zview軟件獲得了奶樣電阻RS、電雙層電容Cdl-T、Cdl-P等電參數(shù),并分析了不同患病程度奶牛奶樣的電參數(shù)特點(diǎn)。最后以鏡檢體細(xì)胞計數(shù)結(jié)果為標(biāo)準(zhǔn),用電參數(shù)作為輸入,建立了牛奶體細(xì)胞數(shù)的支持向量回歸(SVR)預(yù)測模型,研究了體細(xì)胞數(shù)定量檢測的可行性。結(jié)果表明,隨著體細(xì)胞數(shù)的增加(即患病程度的增加),參數(shù)RS減小,而參數(shù)Cdl-T、Cdl-P呈非線性變化趨勢。該定量預(yù)測模型對除N級以外所有奶樣的體細(xì)胞數(shù)都有較高的預(yù)測精度,平均相對誤差為29.40%。1級(隱性)、2級(較嚴(yán)重)和3級(嚴(yán)重)乳腺炎的檢出正確率均達(dá)到100%。

    Abstract:

    A method based on electrical parameters and support vector regression for quantitative detection of somatic cell count (SCC) was exploded. Firstly, the SCC of 100 raw milk samples was confirmed by the standard microscope counting with iodine staining. And then, an interdigitated microelectrode was used in impedance measurements of milk samples in the frequency range from 0.01 Hz to 100 kHz. An equivalent electrical circuit of the milk samples was deduced for the measured impedance data. By using of Zview software, the electrical parameters of the equivalent electrical circuit were extracted and the relationships between these electrical parameters and the degree of mastitis were analyzed. At last, based on these electrical parameters and support vector regression (SVR), a quantitative detection model of SCC was established and its feasibility was studied. It was showed that one of the electrical parameters of RS was decreased with increasing of SCC, while other electrical parameters such as Cdl-T and Cdl-P presented a nonlinear trend. The SVR model was good for prediction of SCC with high precision except the negative samples. The average relative error was 29.40%, and the bovine mastitis detection rate for 1 (sub-clinical), 2 (relative serious infection), and 3 (serious infection) level of samples were 100%. 
