    摘要:

    解淀粉芽孢桿菌是一種植物根際促生細(xì)菌。通過響應(yīng)面試驗(yàn)和正交試驗(yàn),對(duì)分離篩選得到的解淀粉芽孢桿菌L—H15的最大活菌數(shù)進(jìn)行了優(yōu)化,確定了發(fā)酵工藝最佳參數(shù)。在黃瓜育苗期施用高活性固態(tài)菌劑,進(jìn)行實(shí)際效果試驗(yàn)驗(yàn)證。結(jié)果表明,在優(yōu)化的發(fā)酵條件下,即由1∶2.8的麥麩和豆粕組成固態(tài)基質(zhì),外加20%的稻谷殼,0.2%乳糖,0.38%大豆蛋白胨,并用0.4mL/g的液料比混合均勻;裝瓶量46%,解淀粉芽孢桿菌L—H15活菌數(shù)高達(dá)1.68×1010CFU/g;L—H15固態(tài)發(fā)酵菌劑能明顯促進(jìn)黃瓜幼苗的莖粗、株高、葉面積及生物量的累積(P<0.05),根系活力增強(qiáng),礦質(zhì)元素含量也有顯著性提高。因此,解淀粉芽孢桿菌L—H15能夠有效促進(jìn)植株生長,固態(tài)菌劑的研發(fā)為減少農(nóng)藥化肥的施用提供了技術(shù)支撐,是一種提高農(nóng)作物產(chǎn)量的新型方法。

    Abstract:

    Bacillus amyloliquefaciens is a kind of plant growth promoting rhizobacteria. The response surface experiment and orthogonal experiment were used to optimize the number of trophosome of B. amyloliquefaciens L—H15, and determine the optimal conditions of its solid-state fermentation. In addition, B. amyloliquefaciens L—H15 solid-state fermentation inoculant was applied to cucumber plug seedling in order to verify its actual growth-promoting. The optimized fermentation conditions were identified. The solid matrix was composed of wheat bran and soybean (1∶2.8) with external 20% rice hulls, 0.2% lactose and 0.38% soy peptone. And the ratio of water to matrix was 0.4mL/g, with 46% bottling capacity, and 5% inoculation (108CFU/mL). Under this condition, the number of B. amyloliquefaciens L—H15 trophosome was as high as 1.68×1010CFU/g. L—H15 solid-state fermentation inoculant could remarkably increase the stem diameter, height, leaf area and accumulation of biomass (P<0.05), and significantly enhance the mineral elements contents. The cucumber root activity was increased by 5.86 times than that without L—H15 solid-state fermentation inoculant. Thus, B. amyloliquefaciens L—H15 can effectively stimulate plant growth, and L—H15 solid-state fermentation inoculant had a good prospect in the field of microbial fertilizer. This means that L—H15 is a good candidate of biofertilizer and biological control agent in the future. The application of this kind of solid fermentation inoculant can provide a new way for improving the yield of agricultural products.
